    Default Keywords in Domains

Do keywords in domains need to be separated with hyphens in order for search engines to read them?

No, but the domain isn't a place to put keywords anyway. In fact, the only place you should be putting words related to your content is in your content.

As far as I know you should put your keywords on the header and you should separate them with comas.

If you need to put keywords in obscure <meta> tags in the header, you're not being explicit enough in your content. Most search engines these days ignore <meta> tags (when classifying pages) anyway.
